Many "locked" characters are earned by achieving specific combat milestones. For example, some characters unlock after you land a finisher (activated when an opponent is below 21 health) 20 times on non-NPC players.
Since many unlocks depend on kills, always aim for opponents with low health to trigger your finisher.
